The liver is a crucial organ for the maintenance of gastrointestinal homeostasis and body function in general. In addition to its role in the digestive process, it plays a central role in transforming and clearing chemicals and is susceptible to the toxicity from these agents. Certain medicinal agents when taken in overdoses and sometimes even when introduced within therapeutic ranges may injure the liver and this chemical-driven liver damage is termed as hepatotoxicity. Liver injury due to prescription and non prescription medication is a growing medical, scientific and public health problem. Worldwide the estimated annual incidence rate of drug induced liver injury is 13.9-24.0 per 100000 inhabitants. Therefore a more objective and reproducible remedy is needed for the management of the crisis.
In Ayurveda clinical features resembling toxicity of different organ systems are dealt in many contexts. Gara visha is a vast concept mentioned in the Ayurvedic classics, which can be comparable with various sources of exposure of toxins in our daily life activities. Any form of incompatible drug combination or less potent toxins that may get into our body accidently or intentionally, will definitely harm the consumers. The concept of gara in Ayurveda holds well worse with the pathology of hepatotoxicity which need to be unfolded in the lights of our holistic approach. Several formulations are also mentioned in Ayurveda classics which are effective in managing toxicity including hepatotoxicity. Visaghna Mahakashaya has been mentioned in Charaka Samhita Sutra sthana 4th chapter Shadvirechanashathasritheeyam as a potent remedy for all toxins. It is selected for assessing the hepatoprotective action in paracetamol induced hepatic cells. Healthy HepG2 cells will be collected from NCCS Pune, cultured appropriately and hepatotoxicity will be induced with paracetamol. MTT, ROS and Annexin V / FITC Assays are carried out in controls and sample added specimens and observations will be statistically analysed.
This Invitro study of Visaghna Mahakashaya will help us to decipher the mode of action of this combination at the hepatocellular level, thereby exploring its scope for further clinical trials. Moreover all the drugs mentioned in this Mahakashaya is locally available and cost effective compared to the existing therapeutic measures.

ABSTRACT
Virudhahara is a unique concept described in Ayurveda. It is said to be the cause of many diseases. Ayurveda clearly defines that certain diet and its combination, which interrupts the metabolism of tissues, which inhibits the process of formation of tissues and which have the opposite property to the tissues are called as virudha. In the current scenario, junk food consumption is increasing day by day. Owing to its nature, property and composition, it can be classified under the concept of virudhahara. Presently, IT field is one of the best career options among young job seekers. As they contribute exponentially for the growth of our country’s economy, it is imperative that we address their health concern. The purpose of the study is to know how much is the prevalence of virudhahara consumption among IT professionals, and if present to know the reason for their increased consumption of virudhahara, and to analyze the awareness level regarding the incidence of health problems due to virudhahara consumption.
The study design is cross sectional survey and the study setting is Technopark, Thiruvananthapuram. Simple random sampling will be done to select the sample. From the population, 381 people will be randomly selected from both genders as per inclusion and exclusion criteria. Data will be collected through combined and modified version of semi- structured, self administrated questionnaire. The collected data will be processed and conclusions will be made accordingly.
