ABSTRACT

In the current scenario, incidence of skin diseases is increasing irrespective of age and sex. Skin being an index of one’s physical and mental health, any diseases affecting it will be a serious concern for that person. As skin is an important interface between man and his environment, any external factors that affect human body has a primary contact with the skin itself. Contact Dermatitis is an inflammatory eczematous skin disease caused by chemicals or metal ions that exert toxic effects without inducing a T-cell response (contact irritants) or by small reactive chemicals that modify proteins and induce innate and adaptive immune responses (contact allergens). Allergic contact dermatitis is a delayed (type IV) hypersensitivity reaction to exogenous contact antigens. Immunological responses produced in this are due to the interaction of cytokines and T cells. Common etiological allergens for Allergic contact dermatitis are nickel, chromium, formaldehyde, fragrance mix, poison Ivy, poison oak, rubber, cosmetics, fabrics and clothings, detergents, solvents etc. Use of Steroids is found to be the major Treatment in this condition.  In Ayurveda, signs and symptoms of contact with poisoned articles and food substances mentioned by Acharya Susrutha are strikingly similar to Contact Dermatitis. VishaghnaDasaimani is a group of 10 drugs enlisted by Acharya Charaka in his treatise CharakaSamhitha . As it is assumed that Allergic Contact Dermatitis can be discussed under the topic Visha in Ayurveda, VishaghnaDasaimani is taken for Lepa and Kshalana in this study to evaluate its action on the same disease. Along with this, PatoladiGana Kashaya, mentioned by Acharya Vagbhata in his treatise Ashtanga Hrdaya is advised for oral administration, as it is also having  vishahara property. To evaluate the effect of VishaghnaDasaimanigana along with PatoladiGana Kashaya in Allergic Contact Dermatitis, a sample of 30 participants with age group between 18-70 years, fulfilling the inclusion- exclusion criteria and assessment criteria arriving at OPD of Dept. of Agadatantra, Govt. Ayurveda College, Thiruvananthapuram are being selected. Trial drug is being given for a period of 14 days. Data will be collected before and after treatment, scored and statistically analysed using ANOVA.

                ABSTRACT

Man is constantly exposed to different kinds of toxins every day. The drugs that we used to treat various diseases may produce toxic side effects in the body. Undesirable activity of a drug molecule or its reactive metabolites other than its target organ is the main reason for drug induced toxicity which limits the usefulness of widely used therapeutic drugs. Cancer is a major health issue and a leading cause of death worldwide. Chemotherapy is most often used in the treatment of cancer. These drugs travel throughout the body destroy the cancerous cells, but also affect normal healthy tissues. Cyclophosphamide is a drug of choice in chemotherapy which produce hemotoxicity as its limiting side effect. Vishayashtyahwayadi yoga is a formulation mentioned under Raktapitta chikitsa in Vishopayogeeya adhyaya of Astanga Samgraha UttaraSthana. This study is an attempt to assess the hemoprotective effect of the above-mentioned formulation. If the formulation is found to be hemoprotective it can be used for addressing the hemotoxicity caused by Cyclophosphamide and such other agents. The formulation consists of 5 ingredients and is given along with the adjuvant Tandulodaka.   Pharmacognostic and Physico-chemical analysis of the formulation will be conducted before the commencement of experimental study. The experimental study will be conducted at Amala Cancer Research Centre, Thrissur for a period of 3 months. 42 Male balb/c mice weighing 25-30gm will be divided in to 7 groups having 6 animals each. Among this, one group will be selected for acute toxicity study and the remaining groups includes normal, Cyclophosphamide alone, Vishayashtyahwayadi yoga at low, medium and high doses along with Cyclophosphamide respectively and vehicle control along with Cyclophosphamide. All the groups will receive their respective drugs for 7 days except the normal group. Changes in the animals will be noted daily and will be sacrificed on 8th day. Cyclophosphamide induced hemotoxicity will be evaluated by biochemical parameters of blood, histopathological examination of liver and bonemarrow cellularity along with Alpha esterase activity. Results will be statistically analysed by ANOVA, followed by relevant post-hoc test. If the results are found to be significant, the formulation can be further studied as a hemoprotective agent against those drugs causing hemotoxicity.
