Regeneration is a remarkable intrinsic capacity of the human body to restore damaged or lost tissues and functions, thereby maintaining homeostasis and overall health. At the level of the organism, the body has a propensity to go through and acquire changes throughout time, which are typically degenerative. Degeneration, often associated with the gradual deterioration of tissues and organs, is a natural consequence of ageing, and a range of genetic, environmental, and lifestyle factors can further exacerbate it. The intricate balance between degeneration and regeneration processes within the human body in biomedical sciences represents a multifaceted and critical study area. Regenerative medicine focuses on the functional restoration of specific tissues or organs in patients with catastrophic injuries or chronic disease states when their bodies natural regeneration processes fall short of what is required. Enriched with tailored approaches to health and disease, in addition to knowledge of distinctive practices using medicinal plants and peculiar dietary regiments, healthcare systems such as Ayurveda can provide a secure alternative to chronic and degenerative diseases and are increasingly used by individuals with such circumstances. Research opportunities for the biology of regeneration are made possible by the trophorestoration property of the medications and therapies indicated under Ayurvedic Rasayanatantra. Since, zebrafish fins make an excellent study tool for cellular and molecular mechanisms of regeneration, this study aimed to evaluate the regenerative potential of Swadamstradi Rasayana, Amalakyadi Rasayana and their ingredients in the amputated caudal fin of zebrafish. The experimental design was constituted by forty-nine fishes distributed into seven aquaria, receiving six treatments and the seventh one as the control group. Fishes from each treatment were anesthetized and had a portion of their caudal fin amputated with the intention to study the caudal fin's regeneration rates and period. Each experimental setup involved measuring each fish separately before and after amputation, followed by allowing the fish to grow while receiving treatment. To determine the regenerated portions, fishes of each group were subjected to image analysis on days 0, 3, 6, 9, 12 and 15

post amputation. The difference between the control group and the treatment exposed groups in the growth parameters such as caudal fin length increment and length gain percent were analysed. The statistical analysis of the experimental data revealed the variation in treatment means, which were then determined and statistically represented by repeated measures ANOVA, post hoc multiple comparison test (LSD) and independent sample t test. The result showed that the fishes in each treatment group reverted back to its pre amputated caudal fin length in shorter growth period compared to the control group (p<.01). The percentage mean caudal fin length increment was significantly different from control in Amalakyadi Rasayana and Amalaki (p<.05). The results of the study demonstrate the significant potential of two Rasayana yoga Amalakyadi Rasayana and Swadamstradi Rasayana, along with their constituent ingredients Amalaki, Tila, Swadamstra and Amruth, in promoting tissue regeneration in the amputated caudal fin of zebrafish. The knowledge could have implications for treating degenerative diseases in humans and could be applied to promote gene therapies or pharmaceutical interventions to promote regeneration

Most of the Ayurvedic formulations are polyherbal in nature and phytoconstituents present in them are responsible for their therapeutic activities. But nowadays herbal drug pharmaceutical industries face many threats and challenges such as scarcity of raw drug materials, substitution and adulteration, reduction in quality, and increase in price of raw materials. Hence in this scenario, maximum utilisation of raw drugs should be ensured while preparing the Ayurvedic medicines for the preservation of biodiversity. The bioactive principles from herbal medicines are extracted with water, oil, ghee, milk, etc. but recovery of active principles into the extracted phase will not be complete. Therefore, the retentate or spent material of Ayurvedic formulations will be a very rich source of bioactive phytocompounds. This research study was conducted to determine the phytoconstituents retained in the spent material of Guduchyadi Kashaya. Physicochemical and phytochemical comparisons were done between Guduchyadi kashaya and subsequent kashayas prepared from its retentate. For this purpose, physicochemical analysis, chromatographic and spectroscopic techniques, mainly HPLC, LCMS/MS, NMR spectroscopy, TPC, and TFC analysis, were carried out in six samples of methanolic extract of lyophilized Guduchyadi Kashaya. As per the findings of this research study, phytoconstituents are retained in the subsequent kashayas prepared from the retentate of Guduchyadi Kashaya.The analysis using TPC, TFC, and LC-MS/MS showed that the concentration of certain phenolic and flavonoid compounds was increased in the subsequent kashayas. Statistically significant increase of berberine content in subsequent kashayas was observed in HPLC analysis. So, it can be concluded that retentate of Guduchyadi Kashaya contain phytoconstituents with significant therapeutic potential.

ABSTRACT

Polycystic Ovarian Syndrome is one of the most common metabolic &reproductive disorder among women of reproductive age. Changes in the lifestyle, diet, stress of modern society leads to increase in prevalence of PCOS upto 20-30%. PCOS effects the richness of feminity as infertility stands out as one of its major complications. Oligomenorrhoea is one among the most common presenting complaints in participants diagnosed with PCOS. Although no direct correlation of PCOS is found in ayurvedic classical texts. Etiopathogenesis of nastharthava, arhavakshaya, and vandhya yoni vyapath mentioned in susrutha samhitha can be considered here. Pushpagni jataharini mentioned in Kasyapa samhitha bears some similarity with this disease. Here the combined effect of Rajapravartini vati & chiruvilwadi kashaya in Oligomenorrhoea associated with PCOS were evaluated. The study drug Rajapravartini vati is mentioned under streerogadhikara in bhaishajyaratnavali and Chiruvilwadi kashaya is mentioned under arsas prakaranam in sahasrayogam. The study design is pre and post interventional study with a sample size of 30. Females in age group of 18-40 years diagnosed as having Oligomenorrhoea associated with PCOS as per Rotterdam criteria 2003, satisfying inclusion and exclusion criteria attending the OPD of Dept of Prasutitantra and streeroga, Govt Ayurveda College, Thiruvananthapuram were selected for the study. Their symptoms were assessed before starting the treatment using a case proforma, USG and necessary lab investigations. In this study 125 mg of Rajapravartini vati followed by intake of 48 ml of Chiruvilwadi kashaya twice daily before food were adviced to the participants for 90 days. Administration of drug starts on the 1st day of visit and continued till the onset of next menstrual cycle excluding the 3 days of menstruation for three consecutive cycles. Follow up without medicines was done for next 3 consecutive cycles. Data obtained was analysed statistically. The study drug was found to be effective in normalizing menstrual interval, reducing the ovarian volume and reducing the BMI, without causing changes in duration of menstruation, amount of bleeding and haemoglobin level with in the study period.

ABSTRACT
Myocardial Reperfusion Injury refers to damage of cardiac tissue due to restoration of blood to an oxygen deprived myocardium. This may trigger series of cascades such as inflammation and oxidative stress induced oxidative damage that can induce secondary cardiac dysfunction, including myocardial stunning, no-reflow phenomenon, reperfusion arrhythmias, and lethal reperfusion injury which can paradoxically reduce the beneficial effects of myocardial reperfusion and lead to lethal damage to myocardium by the triggering of apoptosis. This can be tackled with antioxidants. Brahma Rasayana being one such formulation, which is thought to bring a positive result in the adverse effects of reperfusion injuries owing to the presence of antioxidants as ingredients.

The main objective of the study was to evaluate the activity of Brahma Rasayana in improving the regenerative potential in Myocardial Reperfusion Injury through invitro study in H9C2 cell line. The protocols adopted were estimation of antioxidant activity, preconditioning and postconditioning to know the response to addition of ischemic buffer, Mitochondrial Membrane Potential (MMP) and Reactive Oxygen Species (ROS). DPPH scavenging assay was chosen for estimation of antioxidant activity. On DPPH free radical scavenging assay, maximum antioxidant activity was observed at the concentration 200 μg/ml with percentage inhibition 65.4592%. In preconditioning, Brahma Rasayana provided maximum average cell viability of 62.81 % at a concentration 25 μg/ml. In postconditioning, the maximum percentage viability was 93.01% obtained at 100 μg/ml. Also, Brahma Rasayana showed reduction in mitochondrial depolarization and ROS production.

ABSTRACT

According to American Diabetes Association, Diabetic peripheral neuropathy (DPN) is ‘’the presence of symptoms or signs of peripheral nerve dysfunction in people with diabetes after exclusion of other causes”. DPN is the most common complication of diabetes mellitus. Globally diabetic neuropathy affects approximately 132 million people (1.9% of the population). DPN is by far the most common of all the diabetic neuropathies. Despite efforts to make an early diagnosis and to halt the progression of diabetic neuropathy, currently there is no effective treatment available at a global level.

       In Ayurveda, a clear-cut clinical entity similar to DPN is not seen. When we look in to both, DPN may be considered as later stages of Prameha. Micro and macro vascular changes and neuronal ischemia are the main pathology in diabetic neuropathy which are similar to the rakta dushti and vatadushti .  Here abnormal neurological manifestations are associated with Prameha, a santarpanotha vyadhi, indicate the involvement of avarana pathology. So the study was undertaken in an effort to find an effective Ayurvedic treatment protocol to manage DPN and to provide a better quality of life. It included Deepana Pachana with Vachadi choornam and Nisakathakadi kashayam, Udwartanam with Kolakulathadi choornam, Vicharana snehapanam with Madhuyastyadi tailam in Nisakathakadi kashayam , Abhyanga with Bala tailam and  Usnambu snana , Virechana with Nimbamritaderanda tailam   followed by internal administration of  Vara, Vidanga, Krishna choornam with tailam, ghrita and madhu . Reduction of TCNS score and improvement in   Vibration Perception Threshold (VPT) assessed using Neuropathy Analyser was statistically analyzed and concluded. Statistical analysis showed that there was significant reduction of score in TCNS scale and improvement in VPT assessed through Neuropathy analyzer. Thus the study was concluded to be effective in reducing the signs and symptoms of DPN.

ABSTRACT

Musta is a rhizomatous perennial herb that grows throughout the country, commonly found in waste grounds, gardens, and roadsides. The rhizome/tuber of Musta is a crucial ingredient in Ayurvedic formulations like Mustakadikwatha, Mustakarishta, Asokarishta etc, offering effective remedies for various ailments such as Jwara, Agnimandya, Ajirna, Swasakasa, Atisara, and more. The Ayurvedic Pharmacopoeia of India has identified the source plant of Musta as Cyperus rotundus Linn., belonging to the Cyperaceae family. Though Cyperus rotundus Linn. is widely present in Kerala, it is not extensively cultivated here. Additionally, there is a notable variation in the market price of Musta. This situation raises concerns about possible adulteration or substitution with lower-quality drugs or those from the same family, which can compromise the safety and efficacy of Ayurvedic formulations. To address these concerns, this study was conducted to determine the authenticity of Musta (Cyperus rotundus Linn.) used in Kerala. For this purpose, three sets of samples were collected from raw drug markets in each zone of Kerala North, Central, and South. Each set of sample included one from a rural market, one from an urban market, and one from a manufacturing unit. These samples were compared with genuine Musta and the standards outlined in the API (Ayurvedic Pharmacopoeia of India) through pharmacognostical and various physicochemical evaluations. Organoleptic evaluation revealed slight differences among the samples in terms of size, shape, external surface and internal colour. Results of phytochemical evaluation indicated marked differences between the market samples and the genuine sample in terms of foreign matter, volatile oil content, ash values, extractive values and fibre contents. Qualitative analysis also showed variations in the presence of steroids, phenols, glycosides, tannin and flavonoids among the samples. High-Performance Thin-Layer Chromatography (HPTLC) analysis revealed differences in peaks among the samples. All heavy metals screened were within permissible limits in all samples. Microbial examinations detected presence of mixed colonies of E. coli, Bacillus, and Klebsiella in all samples, and fungal growth of Aspergillus niger in two market samples. Overall, the study findings suggest that Musta available in herbal drug markets in Kerala are genuine but are of inferior quality.
