DISSERTATIONTITLE:EFFICACY OF DRAKSHADI AGADA VERSUS NIMBAPATRA AGADA IN CISPLATIN INDUCED NEPHROTOXICITY IN WISTAR RATS

                                                        ABSTRACT

Drug induced toxicity is a major health problem in the current scenario which limits the usefulness of widely used drugs. Cisplatin is one among the widely used chemo therapeutic agent which causes nephrotoxicity in approximately one third of the patients receiving it. Through this study we are aiming to provide relief to the patients who are on Cisplatin medication. For this, Drakshadi agada mentioned under the treatment of Mandali visha inSusruta Samhita

Kalpasthana Sarpadashtachikitsavijananeeyam chapter is selected. Mandali visha can be corelated with Viper envenomation by its signs and symptoms and mode of death. Viper venom is chiefly hemotoxic and death may occur due to acute renal failure. Therefore, the treatment modalities may target circulatory and renal system. Hence this study aims to prove the nephroprotective effect of Drakshadi agada to tackle the after effects of xenobiotics causing nephrotoxicity. This study also aims to compare the nephroprotective effect of Drakshadi agada with Nimbapatra agada which had proven nephroprotective activity. Physico-chemical evaluation of the formulation will be conducted at Drug Standardization Unit (DSU) Govt. Ayurveda College, Thiruvananthapuram. Before conducting clinical trial on humans, an animal experimental study is planned to conduct at Amala Cancer Research Centre, Thrissur for a period of 3 months. Wistar rats procured from authentic breeding centre will be divided into 7 groups of 6 animals each. The groups designed for the study are; normal, Cisplatin (10mg/kg body weight), Vehicle control along with Cisplatin, Drakshadi agada at high, medium and low doses along with Cisplatin (10mg/kg body weight), Nimbapatra agada standard dose along with Cisplatin. All the groups will receive their respective drugs for 14 days, except Cisplatin which is administered on 7th day. Changes in every animal will be observed each day and animals will be sacrificed at the end of 16th day. Cisplatin induced nephrotoxicity will be evaluated by histological examinations and biochemical analysis of kidney functions. Results will be statistically analyzed by ANOVA, followed by relevant post-hoc test. If the results are significant, the formulation Drakshadi agada can be further studied as a nephroprotective agent against those drugs which cause nephrotoxicity

DISSERTATIONTITLE:AN EXPERIMENTAL STUDY TO EVALUATE THE ANTIBACTERIAL AND ANTIFUNGAL ACTIVITY OF FUMIGATION WITH IKSHUSARJARASADI YOGA

                                                        ABSTRACT

The atmosphere is a complex dynamic natural gaseous system that is essential to support life on earth. But now a days this gaseous balance is disturbed by pollutants particularly bioaerosols originating from bacteria, fungi, virus and parasites which is hazardous to human health as they pose an ability to remain suspended in air for an extended period of time. Health care settings face direct and inevitable threat from infectious bioaerosols. According to WHO at any given time over 1.4 million people across the globe suffer from a hospital acquired infection.The ancient literature of Ayurveda has given evident knowledge regarding the concept of dushita vayu and its ill effects. As a solution to dushita vayu Ayurveda mentions many dhoopana formulations in various classical texts. Dhoopana is a natural and biocompatible method of air purification which causes no harm to the environment unlike various chemicals and aerosols. In this study, the Ikshusarjarasadi dhoopa yoga mentioned in Visharogadhikara of Vangasena samhitha is analysed for its antimicrobial property. The experimental study will be conducted at Government Ayurveda College Thiruvananthapuram and microbiological laboratory of Emmanuel College Vazhichal. Settle plate method and swab test technique will be adopted to collect microorganism from the OPD. Collected bacterial and fungal colonies will be cultured and will be identified the genus based on the morphological character of the colonies. Fumigation of the bacterial and fungal plates will be done with in a glass chamber keeping a same set of control plates without fumigation. Repeated trials of fumigation are done by varying the concentration of medicine and time of fumigation. Effect of dhoopa will be assessed by comparing the number ofcolony forming unit of bacteria in experimental group with the control group and also by comparing the colony diameter of fungi in experimental group with the control group. The ingredients of Ikshusarjarasadi yoga are easily available and since fumigation is an easymethod this dhoopa can be used easily and successfully.
